چکیده

Background In cancer cells, transcriptional gene silencing has been associated with genetic and epigenetic defects. The disruption of DNA methylation patterns and covalent histone marks has been associated with cancer development. In non-neoplastic cells the multifunctional CCCTC-binding factor (CTCF) can serve as a barrier against the spread of DNA methylation and histone repressive marks into promoter regions of tumor suppressor genes such as BRCA1, Rb, p16 and p53. The absence of CTCF has been related with its epigenetic gene silencing. Until recently, microRNA (miRNA) gene silencing was not well understood. In particular, miR125b1 has been suggested to be a miRNA with tumor suppressor activity, and it has been shown to be deregulated in various human cancers.

Role of CTCF in the regulation of microRNA expression

MicroRNAs (miRNAs) are small non-coding RNAs that regulate expression of various target genes. miRNAs are expressed in a tissue-specific manner and play important roles in cell proliferation, apoptosis, and differentiation. Epigenetic alterations such as DNA methylation and histone modification are essential for chromatin remodeling and regulation of gene expression including miRNAs. The CCCTC-...

Cytotoxicity and Apoptotic Effect of Nanoliposomes loaded with curcumin and miR-34a on MCF-7 and HT29 Cell Lines

Background and aims Colorectal and breast cancers are among the most important causes of mortality in the world. In spite of several attempts, their treatment is yet faced with several problems. Therefore, identification of novel therapeutic approaches is needed. Anti-cancer effects of curcumin and miR-34a have been proposed in various studies. Regarding their pro-apoptotic effects in cancer c...

Evaluation of miR-34a Effect on CCND1 mRNA Level and Sensitization of Breast Cancer Cell Lines to Paclitaxel

Background: A growing body of literature has revealed the effective role of miR-34a, as a tumor suppressor and regulator of expression of multiple targets in tumorigenesis and cancer progression. This study aimed at evaluating the potential effects of miR-34a alone or in combination with paclitaxel on breast cancer cells. Methods: After miR-34a transduction by lentiviral vectors in two MCF-7 an...

I-44: Long-Term Sequelae of Polycystic OvarySyndrome: Gynecological Cancer

Women with polycystic ovary syndrome (PCOS) have been reported to be at increased risk of a number of gynecological neoplasias, including endometrial, breast, and ovarian cancer. The data supporting an increased risk is almost entirely inferential, based primarily on small case series or shared risk factors. One of the difficulties in exploring the association between these cancers and PCOS, is...

MiR-6165 Dysregulation in Breast Cancer and Its Effect on Cell Proliferation and Migration

Background: ncRNAs have been identified as oncogenic drivers and tumor suppressors in any type of cancer. Although many classes of ncRNAs have been reported, most studies have been performed on microRNAs (miRNAs). miRNAs can regulate several target genes and affect important processes such as homeostasis, angiogenesis, cell proliferation, differentiation, and apoptosis. Located in the p75NTR ge...
